Combination therapy with dapoxetine plus sildenafil appears to improve ejaculation time and erection quality in erectile dysfunction (ED) patients with concomitant premature ejaculation (PE), a study has shown.

The study included 53 men (mean age, 45.32 years; mean body mass index, 27.35 kg/m²) with lifelong or acquired PE and ED, all of whom used dapoxetine/sildenafil (30/50 mg film-coated tablet) 1–3 hours before sexual intercourse (2 days a week and no more than once a day) during a 4-week treatment period.

All patients recorded their intravaginal ejaculatory latency time (IELT) with a stopwatch for 4 weeks. They also completed the Premature Ejaculation Diagnostic Tool (PEDT), Premature Ejaculation Profile (PEP) and International Index of Erectile Function-Erectile Function (IIEF-EF) questionnaires before treatment.

By the end of the 4-week treatment period, the mean IELT of the patients substantially increased from 22.72 seconds at baseline to 68.25 seconds (p<0.001). Additionally, there were significant improvements observed in the mean PEP index score (from 0.86 to 2.36; p<0.001) and mean IIEF-EF domain score (from 13.17 to 24.60; p<0.001).

The majority (81.13 percent) of patients were satisfied with the treatment, based on the global impression of change assessment.

In terms of safety, nonserious adverse events (AEs) occurred in 10 patients (18.87 percent), among whom four (7.55 percent) discontinued treatment. Headache, palpitation and flushing were the most commonly reported AEs.

According to researchers, the study is the first to demonstrate the efficacy and safety of oral tablets containing dapoxetine and sildenafil for treating concurrent PE and ED. These tablets can facilitate treatment of prevalent sexual problems by improving both the ejaculation time and erection quality.
